Concerns have been raised over a potential link between thyroid cancer and a class of weight-loss medications known as GLP-1 receptor agonists, which include widely used drugs such as Ozempic, Wegovy, and Mounjaro. These medications have gained popularity for their effectiveness in treating obesity and reducing risks of cardiovascular disease and kidney failure, but questions about their long-term safety have surfaced amid reports of thyroid tumors in animal studies.
GLP-1 receptor agonists work by mimicking a hormone that regulates appetite and blood sugar, promoting significant weight loss. However, early preclinical studies conducted in rodents found that some of these drugs caused thyroid C-cell tumors. Although large-scale clinical trials involving tens of thousands of human patients have not demonstrated a similar cancer risk, regulatory safety information included warnings about these animal findings. For semaglutide—the active component in Ozempic and Wegovy—official prescribing information notes that the relevance of rodent thyroid tumor data to humans is considered low but cannot be entirely excluded. Tirzepatide, the active ingredient in Mounjaro, showed comparable effects in rats, though human implications remain unclear.
In recent weeks, online forums have seen multiple personal accounts from patients who reported thyroid cancer diagnoses after using GLP-1 drugs. While these anecdotal reports cannot establish causality and have not been independently verified, they have raised concern among patients and clinicians, particularly as many individuals may require long-term or lifelong treatment.
Medical experts advise precautions for certain populations. The British Thyroid Foundation recommends that people with a personal or family history of medullary thyroid cancer (MTC), a rare cancer originating in thyroid C-cells, avoid these medications. Similarly, individuals with multiple endocrine neoplasia type 2 (MEN2), an inherited syndrome associated with high MTC risk, should refrain from taking GLP-1 agonists. Patients with prior thyroid cancer are also urged to discuss potential risks with their healthcare providers.
GLP-1 receptor agonists have been used for over a decade to treat type 2 diabetes at lower doses. However, doses prescribed for obesity are significantly higher, and long-term safety data at these levels remain limited. According to Professor Penny Ward of King’s College London, no increased cancer risk has been observed so far, but the possibility of risks emerging with prolonged use or higher doses cannot be fully ruled out, especially given the slow-developing nature of many cancers.
Manufacturers maintain that patient safety remains a top priority. A spokesperson for Novo Nordisk, the producer of Ozempic and Wegovy, emphasized the importance of consulting healthcare professionals regarding side effects and treatment suitability. Similarly, Eli Lilly, maker of Mounjaro, stated that current data do not show a causal link between their drug and thyroid cancer in humans, and they continue to monitor safety profiles closely.
As GLP-1 receptor agonists continue to be prescribed widely for weight management, health authorities and researchers remain vigilant, calling for ongoing surveillance and further investigation into the long-term risks associated with these therapies.
