The appeal of longevity has made it a lucrative market, but scientific validation remains limited and complex.
One popular category of longevity supplements centers on molecules related to nicotinamide adenine dinucleotide (NAD), a vital coenzyme found in every living cell that plays a critical role in energy production and DNA repair. NAD itself cannot be consumed directly because it breaks down during digestion. Instead, people take precursors such as nicotinamide mononucleotide (NMN), which the body can convert into NAD. NMN supplements have gained traction partly due to the advocacy of Harvard researcher David Sinclair. In 2003, Sinclair’s study on resveratrol, a compound found in grape skins, suggested it could activate a gene linked to delayed cellular aging. Although initial enthusiasm for resveratrol was high—boosting supplement sales and even red wine consumption—later clinical trials failed to confirm its efficacy, leading to waning scientific interest.
Sinclair later shifted focus to NMN, publicizing his personal regimen of taking 1 gram each morning and claiming notable improvements in his blood markers related to aging. His book published in 2019, *Lifespan: Why We Age – And Why We Don’t Have To*, further propelled NMN’s popularity. Despite Sinclair's public endorsements, no large-scale, well-controlled human studies have definitively demonstrated that NMN or any other supplement extends lifespan in healthy individuals.
The commercial production of NAD precursors was initially industrial rather than medicinal. A Shanghai-based company, SyncoZymes, pioneered a manufacturing process for NAD as a raw ingredient before the anti-aging potential of NMN became widely recognized. The company’s CEO, Dr. Wei Zhu, an organic chemist involved in ongoing research, began taking NMN himself after reviewing the scientific literature on its role in enhancing DNA repair enzymes that decline with age. Wei reported improved sleep quality and increased energy after starting NMN, experiences he has maintained over seven years.
While anecdotal reports like Wei’s suggest possible benefits, experts caution that the science behind longevity supplements is far from conclusive. Researchers acknowledge that the molecular pathways targeted by NAD and NMN are promising, but robust clinical trials are necessary to substantiate claims of extended human lifespan. As the market continues to grow, consumers face the challenge of distinguishing hopeful marketing from rigorous scientific evidence in the evolving field of longevity research.
